چکیده
To halt North Sea ecosystem degradation, accurate and intensive monitoring of the its fish is vital to correctly inform management decisions. DNA based techniques especially use environmental (e)DNA from seawater can become a powerful tool. However, current eDNA metabarcoding approaches are on genetic target regions <500 nucleotides which offers only limited taxonomic resolution. We tested sensitivity applicability for field samples newly designed universal primer targeting 2kb region covering mitochondrial 12S 16S genes in samples. Samples were processed using long read nanopore sequencing combination with consensus builder Decona retrieved identities up 99.9%. test accuracy primer, was analyzed tropical aquarium known species composition bony elasmobranchs. This showed that over 50% present be identified. The majority remaining reads identified as -in present- genera explained by an incomplete reference database aquarium. Primers also applied sea Distinct compositions between different locations could observed consisted ecological relevant shows long-read studies. Incomplete databases currently form main bottleneck further develop high resolution strategy. Nevertheless, this study used obtain information elasmobranch beyond.

Long read nanopore sequencing for detection of and HLA
Haplotypes are often critical for the interpretation of genetic laboratory observations into medically actionable findings. Current massively parallel DNA sequencing technologies produce short sequence reads that are often unable to resolve haplotype information. Phasing short read data typically requires supplemental statistical phasing based on known haplotype structure in the population or p...

The revolution of genome sequencing is continuing after the successful second-generation sequencing (SGS) technology. The third-generation sequencing (TGS) technology, led by Pacific Biosciences (PacBio), is progressing rapidly, moving from a technology once only capable of providing data for small genome analysis, or for performing targeted screening, to one that promises high quality de novo ...

Background: The ability to obtain long read lengths during DNA sequencing has several potentially important practical applications. Especially long read lengths have been reported using the Nanopore sequencing method, currently commercially available from Oxford Nanopore Technologies (ONT). However, early reports have demonstrated only limited levels of combined throughput and sequence accuracy...
متن کاملBiomonitoring of marine vertebrates in Monterey Bay using eDNA metabarcoding
Molecular analysis of environmental DNA (eDNA) can be used to assess vertebrate biodiversity in aquatic systems, but limited work has applied eDNA technologies to marine waters. Further, there is limited understanding of the spatial distribution of vertebrate eDNA in marine waters. Here, we use an eDNA metabarcoding approach to target and amplify a hypervariable region of the mitochondrial 12S ...
